【Description】

• TOROIVD® Gene Test Tube is a POCT brick specially designed for "Dropper PCR", which can be easily set up a qPCR reaction with the Self-Collection Tube and 5G Reaction Mix bricks. During the use process, it does not rely on professional personnel，pipettes and any instruments . The tube contains freeze-dried mixture of reverse transcriptase, RNase Inhibitor, the primer-probe sets of target gene and IC gene. The probe of internal control gene is labeled with ROX and the amplification curve is yellow.The internal control can monitor the whole procedures including reagents, operation and qPCR cycler, to avoid false negative results. The probe of  target gene probe is labeled with VIC, and the amplification curve is green. Using the POCT bricks, target gene of a certain disease can be quickly detected with direct 1-step RT-qPCR technology.

【Usage】

    -Open the reagent packaging and check if the freeze-dried powder in the tube is deliquescent.

     -Cut open the 8-strips tube according to the required quantity.

     -Remove the transparent dropper cap of the Self-Collection Tube with sample.

     -Drop 1 drop sample into the test tube directly above the tube.

      - Fully thawed 5G Reaction Mix to room temperature and remove dropper cap.

       -Drop 1 drop 5G Reaction Mix into the test tube directly above the tube.

  - Please cover the test tube cap and lightly flick the tube wall. Fully mix and place it in the qPCR cycler.

【Results Interpretation】

 1. The prerequisite of interpretation

• -If the yellow amplification curve appears, but the Cq value of IC-ROX is > 40; or no yellow amplification curve appears with no Cq of IC-ROX.The assay run is invalid. Please check the whole procedures including reagents,  operation and qPCR cycler.

     -If the yellow amplification curve appears, and the Cq value of IC-ROX is ≤40. The assay run is valid.

• -In NTCs tests under the valid run, the green amplification curve appears and the Cq value was generated in Target Gene-VIC. The result is false positive caused by contamination.Please remove the contamination from the environment and reagent consumables

     -In NTCs tests under the valid run, no green amplification curve appears with no Cq of Target Gene-VIC.No false positive is caused by contamination.

2. Results interpretation

     -Under the prerequisite of ensuring valid assay runs and no false positives caused by contamination, the results can be interpreted as follows.

        -If the green amplification curve appears,but the Cq value of Target Gene-VIC is > 40; or green amplification curve disappears with no Cq of Target Gene-VIC .The result is negative.

       -If the green amplification curve appears,and the Cq value of Target Gene-VIC is 15 < Cq <35. The result is positive .

      -If the green amplification curve appears,but the Cq value of Target Gene-VIC is Cq ≤15 or 35 ≤Cq≤40. Retest is needed. If the green amplification curve only appears once in the 2 retests, the result is positive.

       If no green amplification curve only appears with no Cq in the 2 retests,the result is negative.

【Components】

     Freeze-dried mixture of Reverse transcriptase, RNase Inhibitor, the primer-probe sets of Target Gene and IC gene in the 0.2 mL 8-Strips qPCR tube.

【Storage】

     Store the test tube at 2-8℃ in a dry environment for 24 months.

     After opening, the remaining test tubes should still be sealed in a dry environment.

     It is not recommended to use moisture absorbing test tube.

       Ruminants Dieases List: about 46 of  pathogenes   for bovines, sheep and goats;

       Poultry  Dieases List: about 57 of  pathogenes   for chickens, duckes and gooses;   

       Crustaceans  Dieases List: about 9 of  pathogenes   for shrimps;

       Pets  Dieases List: about 48 of  pathogenes   for dogs and cats;

      You  can ask for free samples of the test tubes with your interest sequence of  the target pathogen. When our listed sequences cannot meet your needs, you can offer own designed sequence to get a customized test tube, and each sequence would be named one sequence number(S/N).
